Are there specific letters or numbers in the NI that will show the sex of a person?
Do they also give information as to the DOB of a individual?

These days population are allocated a NI number at birth, although they are not notified of it until they are coming up to 16.
Years ago, when I started out, you applied to the local Labour Exchange for your number when you started work. Some people started at 15, others who go to university may have been 20. Numbers be given out in numerical order on application, as a consequence in those days, age had no relevance. I presume the same thing probably applies to immigrant now, although there may be a series of numbers kept especially for them.
National Insurance numbers own never been gender specific, and do not contain date of birth details.
Are you thinking of this? Sometimes, where on earth an employer doesn't know a NI number, they will use a temporary one, example TN010191F, temporary number/date of birth/gender. This however is not representative, and has no significance to HMRC and the Contributions Office. HMRC has a different system of intervening numbers. No, although you can make a good guess at the year a being first registered for NIC, which is normally at 16, by the first two letters. For example, I believe that YP ability people born around 1950.
